What is IGF-1 LR3?

IGF-1 LR3 modifies native human IGF-1 by substituting arginine for glutamic acid at position 3 and adding a 13-residue extension at the N-terminus. These changes were designed specifically to reduce its affinity for the binding proteins that normally regulate native IGF-1 activity.

How it is understood to work

By evading binding proteins, IGF-1 LR3 remains active in solution far longer than native IGF-1, activating the IGF-1 receptor to drive PI3K/Akt and MAPK signaling cascades associated with cell proliferation, differentiation and protein synthesis.

Research status

It is a standard tool in cell-culture research, particularly studies of muscle satellite-cell proliferation, hypertrophy signaling and general growth-factor biology, where its extended activity window makes it useful compared with native IGF-1.
