What is Melanotan 2?

Melanotan 2 is a seven-amino-acid cyclic peptide originally developed at the University of Arizona during sunless-tanning research. It is structurally related to, but pharmacologically broader than, the more receptor-selective compound Melanotan 1, also known as afamelanotide.

How it is understood to work

Unlike Melanotan 1, which acts mainly at MC1R, Melanotan 2 is a non-selective agonist across MC1R, MC3R, MC4R and MC5R. MC1R activation on melanocytes drives melanin production, while central MC3R/MC4R activity in the hypothalamus has been linked in study models to effects on appetite and sexual-arousal pathways, part of why MT-2 shows a broader activity profile than the more selective Melanotan 1.

Research status

Melanotan 2 is studied in research on skin pigmentation, melanocortin receptor pharmacology, appetite regulation and sexual-arousal signaling. Its non-selective receptor activity subsequently informed the development of bremelanotide (PT-141), a related, more targeted melanocortin agonist now FDA-approved for a specific indication, though Melanotan 2 itself remains an unapproved research compound.
