What is IGF-1 LR3?
IGF-1 LR3 modifies native human IGF-1 by substituting arginine for glutamic acid at position 3 and adding a 13-residue extension at the N-terminus. These changes were designed specifically to reduce its affinity for the binding proteins that normally regulate native IGF-1 activity.
How it is understood to work
By evading binding proteins, IGF-1 LR3 remains active in solution far longer than native IGF-1, activating the IGF-1 receptor to drive PI3K/Akt and MAPK signaling cascades associated with cell proliferation, differentiation and protein synthesis.
Research status
It is a standard tool in cell-culture research, particularly studies of muscle satellite-cell proliferation, hypertrophy signaling and general growth-factor biology, where its extended activity window makes it useful compared with native IGF-1. It has not been developed as an approved therapeutic and clinical safety data in humans is limited.

Handling & storage
Supplied as a lyophilized powder; store unopened vials at -20 C protected from light, and after reconstitution with bacteriostatic water keep refrigerated and use within the timeframe set by the study protocol.

Questions
Why use IGF-1 LR3 instead of native IGF-1 in a study?
Native IGF-1 is rapidly bound and cleared from solution, giving it a half-life of minutes; the LR3 modification resists that binding, giving researchers a longer, more stable signaling window for in vitro work.
Does IGF-1 LR3 activate the insulin receptor as well?
At sufficiently high concentrations IGF-1 LR3 can cross-react with the insulin receptor, which is a consideration researchers account for when interpreting metabolic versus growth-signaling data in a given assay.
