What is GHRP-6?

GHRP-6 (His-D-Trp-Ala-Trp-D-Phe-Lys-NH2) was among the earliest synthetic peptides developed to trigger growth-hormone release independently of GHRH. Its discovery helped establish the existence of a separate GH-secretagogue receptor pathway later identified as the ghrelin receptor.

How it is understood to work

GHRP-6 activates the ghrelin receptor (GHS-R1a) on pituitary somatotrophs, prompting growth-hormone release, and this same receptor’s presence in the hypothalamus underlies GHRP-6’s pronounced effect on hunger signaling in study models. Secondary, more modest increases in cortisol and prolactin have also been reported.

Research status

It has been studied extensively in animal and in vitro pituitary models of GH secretion, and in appetite and cytoprotection research, with a smaller body of early human pharmacology work from decades ago; it has not been developed into an approved GH-related therapeutic, and modern GH secretagogue research has largely moved toward newer analogues.
