What is Cagrilintide?

Cagrilintide is a synthetic, lipidated analog of amylin, the peptide hormone that pancreatic beta cells co-release with insulin. Attachment of a fatty-acid side chain allows it to bind circulating albumin, extending its activity well beyond that of native amylin.

How it is understood to work

It is thought to act on amylin and calcitonin receptor complexes in the brainstem, particularly the area postrema, slowing gastric emptying and contributing to a feeling of fullness; these effects are considered complementary to, rather than duplicative of, GLP-1 receptor signaling.

Research status

Cagrilintide is under clinical investigation, most notably in combination with semaglutide, for its effects on body weight and glycemic parameters in obesity and type 2 diabetes research.
