What is PNC-27?

PNC-27 links residues 12-26 of the p53 tumor-suppressor protein, the region that binds the regulatory protein HDM-2, to a membrane-residency peptide sequence. The design is intended to exploit a feature reported in many cancer cell lines: elevated HDM-2 expression on the outer cell membrane, which is largely absent from normal cells.

How it is understood to work

PNC-27 is reported to bind membrane-associated HDM-2 on cancer cells and, through its membrane-penetrating segment, form pores in the cell membrane that cause rapid cell lysis. Because this action is described as necrosis triggered at the membrane rather than classical apoptosis through intracellular p53 signaling, researchers study it as a mechanism distinct from typical p53-pathway reactivation strategies.

Research status

PNC-27 is studied almost exclusively in in-vitro oncology research, using cultured cancer cell lines to examine membrane-targeted cytotoxicity and selectivity relative to normal cells lacking membrane HDM-2.
