What is PE-22-28?
PE-22-28 (Gly-Val-Ser-Trp-Gly-Leu-Arg) corresponds to residues 22-28 of the sortilin propeptide, the precursor from which the endogenous peptide spadin is cleaved. It was identified in a 2017 study that searched for the shortest spadin fragment that kept its biological activity.
How it is understood to work
Spadin and its analogs block TREK-1, a two-pore-domain potassium channel implicated in mood regulation; mice lacking TREK-1 show a depression-resistant phenotype. In HEK cells expressing human TREK-1, PE-22-28 inhibited the channel at sub-nanomolar concentrations, markedly more potently than full-length spadin.
Research status
Published work is preclinical: rodent behavioral models of depression, measures of hippocampal neurogenesis and synaptic markers, and electrophysiology on cloned channels. The analogs were reported to act faster and last longer than spadin in these models, but no human clinical trials have been published.

Questions
How is PE-22-28 related to spadin?
Spadin is a 17-amino-acid peptide released from the sortilin propeptide. PE-22-28 keeps only seven of those residues, which researchers found retained, and in cell assays improved, TREK-1 inhibition.
What should its COA confirm?
An HPLC purity figure and a mass-spec result matching the expected molecular weight for GVSWGLR (about 802 Da), plus the measured content per vial against the labeled amount.
How much PE-22-28 is in this kit?
10 vials of 10 mg each, so 100 mg in total. The lab testing section above shows what the brand’s tested vial actually contained.
