What is PE-22-28?
PE-22-28 (Gly-Val-Ser-Trp-Gly-Leu-Arg) corresponds to residues 22-28 of the sortilin propeptide, the precursor from which the endogenous peptide spadin is cleaved. It was identified in a 2017 study that searched for the shortest spadin fragment that kept its biological activity.
How it is understood to work
Spadin and its analogs block TREK-1, a two-pore-domain potassium channel implicated in mood regulation; mice lacking TREK-1 show a depression-resistant phenotype. In HEK cells expressing human TREK-1, PE-22-28 inhibited the channel at sub-nanomolar concentrations, markedly more potently than full-length spadin.
Research status
Published work is preclinical: rodent behavioral models of depression, measures of hippocampal neurogenesis and synaptic markers, and electrophysiology on cloned channels. The analogs were reported to act faster and last longer than spadin in these models, but no human clinical trials have been published.
