What is Tirzepatide?

Tirzepatide is a 39-amino-acid synthetic peptide based on the native GIP sequence and engineered to also activate the GLP-1 receptor. A C20 fatty-diacid moiety enables albumin binding, giving it a half-life of approximately five days.

Mechanism of Action

By engaging both incretin receptors, tirzepatide enhances glucose-dependent insulin secretion, lowers glucagon and slows gastric emptying. GIP-receptor activity is thought to contribute additional effects on lipid handling and adipose-tissue metabolism beyond GLP-1 agonism alone.

Research Applications

Tirzepatide is investigated in research on type 2 diabetes, obesity, metabolic dysfunction–associated liver disease and cardiometabolic risk, and is frequently used as a comparator to selective GLP-1 agonists in dual-agonist studies.
