What is FOXO4-DRI?

FOXO4-DRI is a synthetic peptide built using D-amino acids in reverse sequence order, a design known as retro-inverso that is intended to resist normal proteolytic breakdown while still binding its target. It was designed specifically to interfere with a protein-protein interaction implicated in why senescent cells resist programmed cell death.

How it is understood to work

In senescent cells, the transcription factor FOXO4 binds and retains p53 in the nucleus, which blocks the apoptosis pathway p53 would otherwise trigger. FOXO4-DRI competitively binds FOXO4, and in preclinical models this releases p53 to the cytoplasm and selectively induces apoptosis in senescent cells while leaving healthy cells comparatively unaffected.

Research status

The senolytic concept behind FOXO4-DRI was described in a widely cited 2017 study that reported improved fitness, coat condition, and kidney function in aged and progeroid mice. Subsequent research has used it in models of chemotherapy-induced senescence and tissue regeneration in aged animals, but this work remains at the animal and cell-culture stage, with no human clinical development reported.
